 fluorescence correlation spectroscopy (FCS) is a quantitative measurement of the resolution and sensitivity of confocal microscopes. In this tutorial we follow molecular brightness, the count rate per molecule per excitation power, as a well suited parameter for comparison of instrument performance and quality of adjustment.
